Biogenèse des Signaux Peptidiques (BIOSIPE)

Les peptides bioactifs (neuropeptides, hormones, etc.) jouent un rôle clé dans plusieurs processus physiologiques et pathologiques. Les peptides (naturels ou dérivés) sont de plus en plus utilisés comme agents thérapeutiques et leur caractérisation structurale et fonctionnelle ouvre la voie à de nouvelles pharmacopées. La peau des amphibiens contient des quantités importantes de peptides qui sont identiques ou analogues à ceux produits chez les Mammifères. Cette chimiothèque naturelle peut être exploitée pour découvrir de nouveaux peptides endogènes d’intérêt pharmacologique ou thérapeutique.

Une des thématiques de recherche développée par l’équipe BIOSIPE utilise la richesse et la diversité du peptidome de la peau d’amphibien dans le but d’identifier de nouveaux effecteurs impliqués dans le système immunitaire inné. Notre équipe a caractérisé plusieurs PAM appartenant à différentes familles (dermaseptines, phylloxines, dermatoxines, plasticines, phylloseptines) et a défini leurs cibles et leurs mécanismes d’action. Actuellement, nous nous intéressons à de petits PAM cationiques (8-17 résidus) de la famille des temporines, qui ont été isolés à partir de la grenouille nord-africaine Pelophylax saharicus. Ces PAM, appelés temporines SH, possèdent un large spectre d’action antimicrobien, incluant des bactéries multi-résistantes et des parasites.

Les temporines servent actuellement d’outils pour analyser le mécanisme d’action antiparasitaire des PAM qui reste aujourd’hui inconnu. Des études de relation structure-activité sont aussi réalisées pour développer une nouvelle classe de médicaments peptidiques qui pourraient être utilisés pour lutter contre les bactéries multi-résistantes, les biofilms et les parasites.
